Race organisers say they have decided to hold the race later than usual this year due to 'signficant' works currently taking place on the route.
In a Facebook post, they added: "One of the reasons is that the Port 10k is later this year is there are significant works on the route at present.
"The advantage is that there will be a brand new 5m wide footpath and cyclepath to run on back from Kingston Dock into the Town Centre.
"There have also been other improvements to the route in Kelburn Park.
"Given the recent improvements to Mirren Shore it has been decided this year to review the route and we hope these changes will make the course even faster."
The run's planned route starts at Coronation Park and follows the Clyde along to Kingston Dock before heading through Port Glasgow Town Centre and on towards Kelburn Park.
From there the race doubles back past Newark Castle before reaching the finish line at Coronation Park.
The race takes place on pavenments and is suitable for anyone fit enhough to run or walk its 10 kilometre distance.
Entrants must be aged 15 or older on the day of the race.

Trophies will be presented for the first male and first female finisher, with medals awarded to all who take part.
